小女子深受luciferase assay的internal control困擾
所以上網找其他人有沒有這類的東西
看到了一個網址

可是這個人的問題是他的internal control會降低
跟我的不太一樣
後面有個人(doles)有建議 可以試試看phRL vector
是個合成的RL, 據說比較不會受到non specific的activator 影響
而且我剛剛又去看了一下珮雯學姊的論文
他用的就是Promega的phRL-TK
是因為這個原因所以他做起來才不會受影響嗎?

ReplyDeleteI thought the difference between phRL-TK and pRL-TK is the renilla protein, not the promoter regions that TF touches!
我剛剛check了一下

1.modified sequence of RL would express more in mammalian cells
2.mammalian transcription factor binding sites on RL were largely removed
